Glucagon-Like Peptide-1 — a gut incretin hormone whose receptor is the shared target of the dual- and triple-agonist research compounds.
GLP-1 (Glucagon-Like Peptide-1) is an incretin: a hormone released by cells in the gut wall in response to food. Its native role is to amplify glucose-dependent insulin release, slow gastric emptying and signal satiety. Because it is degraded within minutes by the enzyme DPP-4, the native hormone itself is impractical as a research tool, and the compounds studied in its place are engineered analogs with far longer half-lives.
A "GLP-1 agonist" is any molecule that activates that receptor. The distinction that matters when reading the literature is how many receptors a compound engages: single-target GLP-1 agonists, dual GIP/GLP-1 agonists such as Tirzepatide, and triple GLP-1/GIP/glucagon agonists such as Retatrutide. GIP is the other principal incretin; glucagon works in the opposite direction on blood glucose but is studied for a role in energy expenditure, which is why a triple agonist is not simply "more of the same".
