Retatrutide (development code LY3437943) is a synthetic peptide agonist at three receptors at once: GLP-1, GIP and glucagon. Tirzepatide, the compound it is most often compared with, acts at the first two. The glucagon arm is what makes Retatrutide a "triple agonist" and is the part of its mechanism still being characterised — it is studied for a contribution to energy expenditure rather than appetite signalling alone.
Its principal published dataset is a Phase 2 trial reported in the New England Journal of Medicine in 2023 (Jastreboff et al., n=338). In the 12 mg arm, mean body weight fell 17.5% at 24 weeks and 24.2% at 48 weeks. That is a BODY-WEIGHT figure; it is frequently and incorrectly quoted as a body-fat figure, which is a different quantity measured a different way.
Evidence on triple agonism is preliminary — Phase 2 is an earlier stage than the completed programme behind Tirzepatide.
