Cagrilintide
Also known as CagriSema (with semaglutide)
A long-acting amylin analogue, studied alone and in a fixed combination with semaglutide (CagriSema).
In 60 seconds
Phase 2 monotherapy data (2021) and phase 3 combination trials with semaglutide (REDEFINE, 2025–2026). Trials report the gastrointestinal profile expected of the incretin/amylin class; the phase 1b study characterised tolerability of the combination.
- A phase 2 dose-finding trial measured cagrilintide’s dose–response on body weight, safety and tolerability.
- A phase 3 trial measured weight change with the cagrilintide–semaglutide combination (CagriSema).
- A phase 3 trial of CagriSema in adults with type 2 diabetes.
Mechanism
Amylin is a pancreatic hormone co-secreted with insulin that induces satiety and slows gastric emptying. Cagrilintide is a lipidated, long-acting analogue acting at amylin (and calcitonin) receptors, complementing GLP-1 receptor agonism.
Research status
Phase 2 monotherapy data (2021) and phase 3 combination trials with semaglutide (REDEFINE, 2025–2026). Not yet approved.
