In 60 seconds

Approved for type 2 diabetes (twice-daily and weekly forms); extensively used in preclinical research including neuro and β-cell studies. Nausea is the characteristic early effect; reviews cover pharmacokinetics, adverse effects and interactions.

  • Reviews exenatide’s pharmacology, pharmacokinetics, efficacy and adverse effects; derived from Gila monster saliva.
  • A development review of exenatide (synthetic exendin-4), a 39-amino-acid GLP-1 agonist.
  • In a mouse xenograft system, exendin-4 combined with harmine expanded human β-cell mass.

Mechanism

Exendin-4 shares about 53% homology with GLP-1 but resists DPP-4 degradation. It is a full GLP-1 receptor agonist, enhancing glucose-dependent insulin release, suppressing glucagon and slowing gastric emptying. It remains a standard research tool for GLP-1 receptor biology.
