Reproductive-axis (HPG) peptides
Kisspeptin-10
Also known as kisspeptin · metastin 45-54 · KP-10
The shortest active fragment of kisspeptin, the hypothalamic peptide that triggers GnRH release and sits at the top of the reproductive axis.
In 60 seconds
Human research tool in reproductive endocrinology (fertility, hypothalamic amenorrhoea); not an approved medicine. Short-term human studies are physiological; repeated exposure can desensitise the axis in some protocols.
- Reviews kisspeptin as the critical central regulator of GnRH release.
- Reviews how metabolic signals act on kisspeptin neurons to link energy balance and fertility.
- Reviews kisspeptin and neurokinin B in puberty, the menstrual cycle and menopause.
Mechanism
Kisspeptin binds its receptor (KISS1R/GPR54) on GnRH neurons, driving GnRH pulses and therefore LH, FSH and testosterone/oestrogen. Kisspeptin neurons integrate metabolic and stress signals, linking energy balance to fertility.
Research status
Human research tool in reproductive endocrinology (fertility, hypothalamic amenorrhoea); not an approved medicine.

Safety signals in the literature
Short-term human studies are physiological; repeated exposure can desensitise the axis in some protocols. A 2024 study also found KISS1R signalling in osteoclasts.
Legal status (South Africa)
Gonadotropins and GnRH analogues are scheduled prescription medicines in South Africa; research use only.
