How to Store Reconstituted Peptides

Once a peptide is in solution the storage rules change completely. Here is exactly how to keep a reconstituted vial stable: temperature, orientation, light, draw discipline and when to discard.

A reconstituted peptide is a far more fragile thing than the freeze-dried powder it came from. The moment water touches the powder, the molecule is exposed to hydrolysis, oxidation and microbial risk, and the generous shelf life of a lyophilised vial collapses to a matter of weeks. Storing the solution well is what protects the money and time already invested in the vial.

This guide is specifically about the reconstituted (in-solution) state — not the sealed powder. If your vial is still lyophilised, the general storage guide covers that. Here we focus on the fridge, why you must never freeze a reconstituted solution, and how your drawing habits affect how long the vial lasts.

STEP-BY-STEP

  1. Refrigerate immediately after mixing

    The instant reconstitution is complete, the vial belongs in the fridge at 2–8°C. Every minute a solution spends warm accelerates degradation. Do not leave a freshly mixed vial on the bench while you tidy up — mix, label, chill.

    Tip: The main body of the fridge is more stable than the door. Keep the vial on a shelf, not in the door rack.

  2. Store the vial upright

    Keep the vial standing vertically so the solution is not in constant contact with the rubber stopper. Prolonged contact with the stopper can, over weeks, allow trace leaching and makes the seal harder to keep clean between draws.

  3. Never freeze a reconstituted solution

    Freezing is for lyophilised powder only. Once a peptide is dissolved, ice-crystal formation and the freeze-thaw cycle physically shear the molecule. A solution that has been frozen and thawed should be treated as compromised.

    Tip: Keep the vial away from the freezer compartment and the coldest back corner of the fridge, where accidental freezing happens.

  4. Keep it dark

    Return the vial to its carton or an opaque container. UV and even strong ambient light degrade peptides in solution over time, and clear glass offers no protection.

  5. Minimise time out of the fridge per draw

    Take the vial out, swab, draw your research measurement, and return it — the whole exercise should take a minute or two. The fewer and shorter the warm excursions, the longer the solution stays good.

  6. Track the mix date and set a discard window

    Write the reconstitution date on the vial and decide, in advance, the date past which you will discard it. Most reconstituted peptides hold for a few weeks refrigerated; committing to a window stops you relying on material that is past its useful life.

  7. Inspect before every draw

    A healthy solution stays perfectly clear. Cloudiness, colour, or floating particles mean it is time to discard. The clarity check is your most reliable in-hand signal of stability.

Why the reconstituted shelf life is so much shorter

Freeze-dried powder has almost no free water, so the chemical reactions that break peptides down are effectively paused. Add bacteriostatic water and those reactions — hydrolysis of the peptide bonds, oxidation of sensitive residues — resume. This is why a vial that was stable for months as a powder is only stable for weeks in solution.

The benzyl alcohol in bacteriostatic water holds microbial growth in check, but it does nothing to stop the chemical degradation of the peptide itself. That is why cold, dark, undisturbed storage matters just as much as sterile technique for a reconstituted vial.
