Insulin Syringe Guide: Reading Units & Gauges

Understand U-100 insulin syringes for research measuring: how units map to millilitres, what gauge and needle length mean, and how to draw an accurate, bubble-free volume.

The insulin syringe is the measuring instrument of peptide research. Its fine barrel and unit markings make it ideal for drawing the small, precise volumes that reconstituted peptides call for. But the units printed on the side confuse a lot of people, because they are not millilitres.

This guide decodes the U-100 scale, explains gauge and needle length, and walks through drawing a clean, accurate measurement.

STEP-BY-STEP

  1. Learn the U-100 scale

    A U-100 insulin syringe is marked in "units", where 100 units = 1 ml. So 10 units = 0.1 ml, and 5 units = 0.05 ml. The calculator converts your draw volume into units automatically, but knowing the relationship lets you sanity-check any number.

    Tip: Units are not millilitres. 1 unit = 0.01 ml on a U-100 syringe.

  2. Pick the right barrel size

    Insulin syringes come in 0.3 ml (30 unit), 0.5 ml (50 unit) and 1 ml (100 unit) barrels. A smaller barrel spreads the markings out, making small draws easier to read accurately. Choose the smallest barrel that still holds your target volume.

  3. Understand gauge and length

    Gauge is needle thickness — a higher number means a thinner needle (e.g. 29G–31G are common and fine for drawing). Length is measured in millimetres. For measuring from a vial, a standard short insulin needle is ideal.

  4. Swab and draw a little air first

    Swab the vial stopper. Pull the plunger to your target volume in air, insert the needle into the upright-then-inverted vial, and push the air in — this equalises pressure and makes the liquid draw smoothly.

  5. Draw to your target line

    With the vial inverted, pull the plunger slowly until the top edge of the plunger seal sits exactly on your target unit mark. Read at eye level to avoid a parallax error.

  6. Clear the bubbles

    Tap the barrel so any air bubbles rise to the top, then push the plunger gently to expel them back into the vial and re-draw to your exact mark. A bubble in the barrel means you draw less liquid than the units suggest.

    Tip: Even a small bubble throws off a small measurement — always clear it before finalising the draw.

Quick unit reference

On a U-100 syringe: 1 ml = 100 units, 0.5 ml = 50 units, 0.1 ml = 10 units, 0.05 ml = 5 units, 0.02 ml = 2 units.

If your target measurement lands on an awkward fraction of a unit, that is a sign your concentration is too high for comfortable measuring — go back to the calculator and reconstitute at a lower concentration (more water) next time so the same amount spreads across more units.
