Angiotensin-(1-7)

Also known as Ang-(1-7) · Ang 1-7 · TXA127

A heptapeptide of the renin–angiotensin system that tends to counter angiotensin II — the "protective arm" of the ACE2/Ang-(1-7)/Mas axis.

Investigational. A phase 1–2 ICU trial reported the infusion as safe, with one serious adverse event (bradycardia) considered possibly related; it showed no significant difference in its phase 2 outcome.

  • A major review of the ACE2/angiotensin-(1-7)/Mas axis and its counter-regulatory role in the renin–angiotensin system.
  • A phase 1–2 randomised trial in ICU COVID-19 patients found the infusion safe but no significant difference in its phase 2 outcome.
  • In a crossover study in mountain bike athletes, oral HPβCD-Ang-(1-7) did not improve time-trial or sprint performance.

Mechanism

ACE2 converts angiotensin II into angiotensin-(1-7), which acts mainly through the Mas receptor. In research models this axis opposes many angiotensin II effects, with vasodilatory, anti-inflammatory and anti-proliferative actions.

Research status

Investigational. Early clinical trials include intravenous infusion in ICU COVID-19 patients and an oral cyclodextrin formulation in athletes; not an approved medicine.
