Pramlintide
Also known as Symlin
A synthetic analogue of the pancreatic hormone amylin, the first approved amylinomimetic.
In 60 seconds
Approved (2005, US) as an adjunct to mealtime insulin in type 1 and type 2 diabetes. Hypoglycaemia risk when combined with insulin is the key labelled concern; a randomised study found it could provoke migraine-like attacks in migraine patients.
- Reviews pramlintide as the first amylinomimetic, approved in 2005 as an insulin adjunct.
- Describes amylin’s glucagon, gastric-emptying and food-intake effects and the clinical data for pramlintide.
- A randomised study in migraine patients found pramlintide induced migraine-like attacks.
Mechanism
Amylin is co-secreted with insulin; it lowers post-meal glucose by suppressing glucagon, slowing gastric emptying and reducing food intake. Pramlintide substitutes prolines to stop native amylin’s tendency to aggregate.
Research status
Approved (2005, US) as an adjunct to mealtime insulin in type 1 and type 2 diabetes. The scientific ancestor of cagrilintide.
