Abaloparatide
Also known as Tymlos · Eladynos · PTHrP(1-34) analogue
A synthetic analogue of parathyroid hormone-related protein (PTHrP 1–34) that selectively activates the PTH1 receptor to build bone.
In 60 seconds
Approved for postmenopausal osteoporosis (prescription-only); the ACTIVE phase 3 trial measured vertebral fractures. Reviews and a 2024 meta-analysis report palpitations, dizziness and hypercalcaemia among adverse events.
- The ACTIVE phase 3 trial measured new vertebral fractures with abaloparatide versus placebo.
- A meta-analysis of abaloparatide’s efficacy and safety in postmenopausal osteoporosis.
- Maps preclinical and clinical knowledge of abaloparatide.
Mechanism
Abaloparatide shares its first 20 amino acids with PTHrP and favours a receptor conformation linked to transient signalling, which is proposed to give a bone-forming effect with less stimulation of resorption than teriparatide.
