The dual GIP/GLP-1 agonist behind SURMOUNT-1 — a −20.9% mean body-weight change at 72 weeks on the 15 mg arm, published in NEJM.

Each vial contains 10mg of lyophilised Tirzepatide, supplied as a sterile powder for reconstitution. Tirzepatide (LY3298176) is a dual incretin receptor agonist developed by Eli Lilly that activates the GIP (glucose-dependent insulinotropic polypeptide) and GLP-1 (glucagon-like peptide-1) receptors simultaneously, which is why it is among the most frequently cited comparators in the recent metabolic literature. In the SURMOUNT-1 trial a mean body-weight reduction of up to 22.5% over 72 weeks was reported in the trial population, and the SURPASS programme reported glycaemic-control endpoints — both figures from those trials, not statements about any individual. Mechanistic work describes engagement of both incretin pathways in relation to insulin secretion, appetite signalling, gastric emptying and insulin sensitivity, and trial programmes have examined hepatic fat content, cardiovascular risk markers and lean mass during caloric deficit.
