5-Amino-1MQ
Also known as 5A1MQ · 5-amino-1-methylquinolinium
A small-molecule inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT), studied in obesity and metabolism models.
In 60 seconds
Preclinical: diet-induced-obese mouse studies (alone and with diet change). No human safety data.
- In diet-induced obese mice, the NNMT inhibitor 5A1MQ was studied for body composition, liver pathology and pharmacokinetics.
- In obese mice, 5-amino-1-methylquinolinium plus a low-fat diet rapidly normalised adiposity and altered the gut microbiome.
- Reviews how NNMT links metabolite levels to chromatin-modifying enzymes and gene regulation.
Mechanism
NNMT methylates nicotinamide using SAM, linking cellular energy (NAD+) balance and methylation capacity. It is over-expressed in adipose tissue in obesity; inhibiting it is proposed to raise NAD+ and shift fat-cell metabolism.
Research status
Preclinical: diet-induced-obese mouse studies (alone and with diet change). No human trials published.

Safety signals in the literature
No human safety data. NNMT is also implicated in cancer biology, an active, separate research area.
Legal status (South Africa)
Approved incretin medicines are prescription-only (scheduled) in South Africa and available only through a prescriber; research-grade material is not a medicine and is not for human use.
