Nicotinamide Riboside (NR)
Also known as NR · Niagen · nicotinamide riboside chloride
A vitamin-B3-form NAD+ precursor that raises blood NAD+ in humans, tested in randomised trials in ageing and neurodegeneration.
In 60 seconds
Multiple randomised trials (safety, Parkinson’s, MCI, COPD, PAD); GRAS status for food use in the US. Long-term trials in overweight adults and a high-dose Parkinson’s safety trial reported it as well tolerated.
- A phase 1 trial tested whether NR is safe and raises cerebral NAD in Parkinson’s disease.
- A randomised safety trial of high-dose NR in Parkinson’s disease.
- A randomised trial assessed long-term NR safety and NAD metabolism in overweight adults.
Mechanism
NR is phosphorylated by NRK enzymes to NMN and then to NAD+. Raising NAD+ is the rationale for trials in Parkinson’s disease, cognitive impairment, COPD and peripheral artery disease.
Research status
Multiple randomised trials (safety, Parkinson’s, MCI, COPD, PAD); GRAS status for food use in the US.
