Mitochondrial & longevity research compounds
NMN (Nicotinamide Mononucleotide)
Also known as β-NMN · beta-nicotinamide mononucleotide
A direct precursor of NAD+, studied in human trials as a way to raise NAD+ levels that decline with age.
In 60 seconds
Multiple small randomised human trials (insulin sensitivity, walking speed, aerobic capacity, sleep). Trials and reviews report it as well tolerated over weeks to months; long-term human data are still limited.
- A randomised trial in prediabetic women measured muscle insulin sensitivity after NMN supplementation.
- A dose-dependent randomised trial assessed blood NAD levels, physical performance and safety of NMN in middle-aged adults.
- In older adults, 12 weeks of NMN raised blood NAD and was associated with maintained walking speed.
Mechanism
NMN is converted to NAD+ by NMNAT enzymes; NAD+ is a cofactor for energy metabolism and for sirtuins and PARPs. Raising NMN availability raises tissue NAD+ in animals, the premise of human supplementation research.
Research status
Multiple small randomised human trials (insulin sensitivity, walking speed, aerobic capacity, sleep). Regulatory status of NMN as a supplement varies by country.
