Orexin-A (Hypocretin-1)
Also known as hypocretin-1 · HCRT1 · OXA
A hypothalamic neuropeptide that stabilises wakefulness; its loss causes narcolepsy type 1.
In 60 seconds
Research peptide; intranasal pilot studies in humans and animal models of cognitive dysfunction. A pilot human study found intranasal orexin A modulated sympathetic vascular tone; no long-term data.
- Reviews orexin-A/B and their two receptors in sleep–wake regulation.
- A randomised pilot study in healthy men measured sympathetic vascular tone after intranasal orexin A.
- In aged rats, intranasal orexin-A affected neuronal activation and attention.
Mechanism
Orexin-A activates OX1R and OX2R receptors, driving arousal, attention and sympathetic tone. Orexin-receptor antagonists are approved for insomnia; intranasal orexin-A is studied as a research tool.
Research status
Research peptide; intranasal pilot studies in humans and animal models of cognitive dysfunction.
