SS-31 (elamipretide) is a cell-permeable tetrapeptide (D-Arg-Dmt-Lys-Phe-NH2) that concentrates in the inner mitochondrial membrane and binds cardiolipin, the phospholipid that organises the electron-transport chain.

  • In an ischaemic kidney model, SS-31 preserved cristae and speeded ATP recovery by interacting with cardiolipin.
  • Reviews elamipretide’s structure, cardiolipin-binding mechanism and research applications.
  • Summarises the September 2025 US accelerated approval of elamipretide for muscle strength in Barth syndrome.

OVERVIEW

SS-31 (D-Arg-Dmt-Lys-Phe-NH2 (elamipretide)) is a research-grade peptide supplied by Living Water Labs for laboratory investigation. Mitochondria-targeted tetrapeptide (elamipretide), 10 mg lyophilised powder per vial.. Each published certificate is an independent Janoshik Analytical report naming the compound and its HPLC purity; certificates not yet published can be requested. The summary below outlines documented mechanisms and study areas reported in the research literature.

DOCUMENTED RESEARCH AREAS

  • ▸Mitochondria-targeted tetrapeptide that concentrates in the inner membrane
  • ▸Binds cardiolipin, the lipid that organises the electron-transport chain
  • ▸Preserved cristae and ATP recovery in ischaemic kidney models

MECHANISM OF ACTION

SS-31 (elamipretide) is a cell-permeable tetrapeptide (D-Arg-Dmt-Lys-Phe-NH2) that concentrates in the inner mitochondrial membrane and binds cardiolipin, the phospholipid that organises the electron-transport chain. In injury models this is reported to stabilise cristae structure and restore ATP production; in 2025 elamipretide received US accelerated approval for muscle strength in Barth syndrome, a rare cardiolipin disorder.

  • ▸Concentrates in the inner mitochondrial membrane
  • ▸Binds cardiolipin, stabilising cristae and electron-transport supercomplexes
  • ▸Restored ATP production in ischaemic kidney models
