What Is Reconstitution?

Reconstitution is the step that turns a freeze-dried peptide powder into a usable solution. It sounds technical — it’s really just careful measuring. Here’s how it works.

The one-line definition

Reconstitution means adding a measured amount of liquid to a freeze-dried peptide powder to turn it into a solution you can accurately measure from. That’s the entire concept — you’re just rehydrating the powder.

Peptides ship as a dry powder because they’re more stable that way; reconstitution is the step that makes them usable in a research context.

What you reconstitute with

The usual liquid is bacteriostatic water — sterile water containing a small amount of benzyl alcohol, which inhibits bacterial growth so a solution stays usable longer than plain sterile water would.

The liquid is added slowly and gently down the side of the vial, never blasted directly onto the powder, because peptides are fragile and rough handling can degrade them.

It’s really just concentration maths

The only "hard" part is arithmetic. Concentration is simply the amount of peptide divided by the volume of liquid. Add 2 ml of water to a 2 mg vial and you get 1 mg per ml. Add 1 ml instead and you get 2 mg per ml — same powder, different concentration.

Choosing the volume up front decides how concentrated every later measurement will be, which is why getting this ratio right matters.
